Support
At V-CEM we offer support in planning of the EM experiments, training in sample preparation and imaging, use of equipment and data analysis. The support can be from full collaboration (you "drop-off" your samples) or, depending on your interest and the nature of your work just help and guidance so that you can do your own work.
All necessary support and assistance are available. :)
Steps to start a new project at the V-CEM:
*** Basic starting point to initiate a project is a biochemically pure sample ***
Sample purity must be improved in each investigator’s laboratory. Some buffers are not compatible with cryoEM and discussion is recommended to build the final-phases of the purification protocol.
- 1. Contact the V-CEM
- 2. Schedule a meeting and discuss goal and a possible plan.
- a. Sample quality (biochemistry, bring your result, structure)
- b. Protein/complex size
- c. Protein/complex stability.
- d. Goal (resolution, negative stain, Cryo)
- 3. Possible tiers to get your project running:
- a. Independent - salf user with staff support (need to be trained and approved user)
- b. Supervised - partial-colabaration data collection
- c. Collaborating - full colabaration data and processing
EM Facility User levels
User levels in V-CEM:
* Independent
* Supervised
* Collaborating users
Independent users
Access: Independent use of the microscope and sample preparation equipment. Access the facility during nights and weekend.
Responsibilities: Complete microscope training (2-3 sessions). After demonstrating your capability on the microscope you will be tested on the microscope and knowledge of our standard operating procedures. You are responsible for all matters relating to your project, but staff will be on hand to assist you set up the microscope and answer general EM questions during normal operation hours.
Acknowledgement: In publications or presentations please note the use of V-CEM.
Supervised users
Access: Supervised use of the microscope and sample preparation equipment during normal hours of operation, pending staff availability.
Responsibilities: Complete microscope training (2-3 sessions). You are responsible for all matters relating to your project, but staff will be on hand to assist you set up the microscope and answer general EM questions. Staff will be available to help with aligning the microscope, sample insertion and optimizing imaging during your session. Outside of your allocated time staff can give you general guidance on your EM project.
Acknowledgement: In publications or presentations please note the use of V-CEM and acknowledge the staff that has assisted you.
Collaborating users
Access: Staff assisted use of the electron microscopy facility for your project.
Responsibilities: Staff will have integral involvement in the EM part of your project to give general guidance and assist in the project from beginning to end. Projects have the most chance of success if there is a dedicated member in the lab devoted to the project.
Acknowledgement: Authorship/co-authorship in publications or presentations commensurate to the contribution.
